transient expression of vp2 gene of very virulent ibdv in tobacco, alfalfa and lettuce leaves by agroinfiltration
Authors
abstract
vp2 protein is the major host protective immunogen of infectious bursal disease virus (ibdv) of chickens. plants are now gaining widespread acceptance as a general platform for the large scale production of recombinant proteins including vaccine, antibodies and pharmaceuticals. in present study vp2 gene of an iranian isolate of ibdv was cloned under strong plant expression promoter camv35s and introduced to agrobacterium tumefaciens. using agro-infiltration method, vp2 protein was expressed in tobacco (nicotiana tobacum), alfalfa (medicago sativa) and lettuce (lactuca sativa) leaves. the expression of vp2 was detected in leaves extracts by western blot and elisa methods. the highest expression was detected in alfalfa and tobacco leaves and lowest expression was found in lettuce.

Transient Expression of Human Growth Hormone in Potato (Solanum tuberosum), Tobacco (Nicotiana tobacum) and Lettuce (Lactuca sativa) Leaves by Agroinfiltration
Using agro-infiltration technique, we have transiently expressed human Growth Hormone (hGH) in tobacco(Nicotiana tobacum), potato (Solanum tuberosum) and lettuce (Lactuca sativa) leaves. Out of three differentinoculation times used for infiltration in our study, it was seen that highest level of hGH expression wasachieved when leaves were infiltrated for 35 min. The presence o...
full textTransient Expression of Foot and Mouth Disease Virus (FMDV) Coat Protein in Tobacco (Nicotiana tabacom) via Agroinfiltration
Background: Transient and stable transformation of host plants are the common techniques to produce transgenic plants. However, the main drawback of stable transformation is the fact that it takes quite a long time to produce a transgenic line. While, transient gene expression is a quick method to produce recombinant proteins in plants. Objective: The main goal of the present study was to eva...
full textA Simple Method for Transient Expression of Reporter Gene in Brinjal Leaves through Agroinfiltration
The soil-borne bacterium, Agrobacterium tumefaciens is called the natural genetic engineer, because it has the capacity to transfer and integrate a part of its tumour inducing (Ti) plasmid DNA (the transfer DNA or T-DNA) into the plant genome. Through exploring this natural system, plant genetic engineers place their gene of interest in the engineered Ti plasmid and deliver the gene of interest...
full textTransient Expression of Biologically Active Anti-rabies Virus Monoclonal Antibody in Tobacco Leaves
Background: Rabies virus is a neurotropic virus that causes fatal, but, a preventable disease in mammals. Administration of rabies immunoglobulin (RIG) is essential for the post-exposure of the prophylaxis to prevent the disease. However, replacement of polyclonal RIGs with alternative monoclonal antibodies (MAbs) that are capable of neutralizing rabies virus has been recommend...

an agrobacterium-mediated transient gene expression assay was carried out in alfalfa (medicago sativa) leaves for expression of a chimeric gene encoding a part of capsid protein of foot and mouth disease virus called vp1. the plant leaves were transformed via agroinfiltration procedure. the presence of the foreign gene and its expression in transformed plants were evaluated by polymerase chain ...
full textLeaf-Encapsulated Vaccines: Agroinfiltration and Transient Expression of the Antigen Staphylococcal Endotoxin B in Radish Leaves
Transgene introgression is a major concern associated with transgenic plant-based vaccines. Agroinfiltration can be used to selectively transform nonreproductive organs and avoid introgression. Here, we introduce a new vaccine modality in which Staphylococcal enterotoxin B (SEB) genes are agroinfiltrated into radishes (Raphanw sativus L.), resulting in transient expression and accumulation of S...
